Description

The INS-1 cell line, derived from x-ray-induced insulinoma in rats, exhibits a high insulin content and responsiveness to glucose changes. Cultivation relies on 2-mercaptoethanol and showcases gradual proliferation (100-hour doubling time) with beta-cell-like morphology. Heterogeneous in nature, INS-1 cells comprise mature insulin-positive and immature bi-hormonal cells co-expressing insulin and glucagon. Bi-hormonal variants, lower in Nkx6.1 and lacking alpha cell markers, imply incomplete maturation. Prolonged glucose exposure diminishes insulin gene and protein expression, subsequently reducing insulin and proglucagon-derived peptide levels, including GLP-1, GLP-2, and glucagon.
