Immortalized Mouse Dendritic Cell Line was isolated from C57BL/6 transgenic mice carrying SV40 Large T oncogene
Immortalized Mouse Dendritic Cells (Mouse DCs) are derived from murine bone marrow and transformed via retroviral transduction of v-raf and v-myc oncogenes to enable continuous proliferation while preserving key dendritic cell functions. The cells retain key functional attributes of dendritic cell lines, providing a stable and renewable in vitromodel for immunological research, where they are often used as a counterpart to primary dendritic cells or other established lines like DC2.4. These cells display characteristic dendritic morphology with cytoplasmic projections and grow adherently or semi-adherently in vitro. They express canonical dendritic markers, including MHC class II, CD11c, CD80, and CD86. Functionally, they retain antigen-presenting capabilities, secrete cytokines such as IL-12 and TNF-α, and effectively stimulate T cells. Offering a scalable and reproducible alternative to primary dendritic cells, they are widely used in studies of antigen processing, T-cell activation, tumor immunology, vaccine development, and autoimmune disease mechanisms.

| Quality Control | 1) Direct antigen presentation and cross-antigen presentation were evaluated by the MHC-I (SIINFEKL/OT-I ) and MHC-II (OVA323-339/OT-II) restricted systems; 3) proteome profile and surface markers assessed by RT-PCR; RT-PCR; 3) IL-12 cytokine secretion analyzed using ELISA, 4) Response to PAMP stimulation evaluated by functional assays. |
Immortalized mouse dendritic cells are widely used in antigen presentation studies (direct and cross-presentation), T-cell activation assays, cancer immunology, vaccine evaluation, and cytokine profiling (including IL-12). Their unlimited proliferative potential allows for consistent and scalable experiments, including gene editing, pathway analysis, and drug screening in immune modulation and autoimmune models.
