MicroRNA Lentivirus Packaging Service
AcceGen has developed the proprietary MicroRNA Lentivirus Packaging Service for research applications. We offer high-quality service for the construction of microRNA overexpression/inhibition lentivector clones and lentivirus packaging. Lentivirus is a genus of retroviruses that expresses reverse transcriptase and integrase. Recombinant lentivector is modified from HIV-1 with the viral genes removed. MicroRNAs are a class of non-coding single-stranded RNA molecules with a length of about 22 nucleotides found in animals, plants, and some viruses. MicroRNAs can regulate the post-transcriptional expression of cognate target genes.
The construction of microRNA overexpression/inhibition lentivector
MicroRNA precursors with their native upstream and downstream flanking genomic sequences are amplified and cloned together into the lentivector. The lentivector containing microRNA precursor is especially valuable for long-term gain-of-function studies. See the figure below for the microRNA precursor structure.
The construction of a MicroRNA sponge vector allows the expression of a transcript known as the microRNA sponge. This sponge comprises multiple tandem high-affinity microRNA binding sites, enabling it to effectively sequester specific microRNAs from their endogenous target mRNAs. This constructed MicroRNA sponge vector is especially valuable for long-term loss-of-function studies. See the figure below for the microRNA sponge structure.
MicroRNA lentivirus packaging
Lentiviral particles are very efficient in delivering microRNA precursors and sponges to a wide range of cell types, even those that are hard to be transfected. They infect cells, leading to the integration of transgenetic information into the host genome, which results in long-term transgene expression. AcceGen has unique expertise in producing, concentrating, and titrating microRNA lentiviral particles, yielding high titers and delivering consistent results.
