293A is a subclone of human embryonic kidney (HEK) 293 cell line, originally derived from fetal kidney fibroblasts transformed with adenovirus type 5 (Ad5). This adherent cell line exhibits epithelial-like morphology with adherent culture properties. These adherent cells exhibit a flat, monolayer growth pattern. 293A retains stable integration of the adenoviral E1 gene, enabling constitutive expression of E1a and E1b proteins, which supports efficient replication of E1-deleted adenoviral vectors. Additionally, 293A expresses an aberrant vitronectin receptor composed of integrin β1 and α-v subunits, enhancing cell adhesion and surface interactions. The cell line’s genetic and morphology features make it particularly valuable for viral plaque assays and adenovirus vector development.

293A cells are a powerful platform for the streamlined production, amplification, and titration of recombinant adenovirus vectors, especially those that are replication-incompetent. Their stable expression of adenoviral E1 proteins supports high-efficiency viral packaging, making them essential for researchers engaged in gene therapy, vaccine development and viral vector engineering. 293A is widely used in transfection assays and is ideal for precise screening and optimization of adenoviral constructs in both biomedical research and preclinical development.
