C3A cell line is a human hepatocyte-derived model supporting liver biology, metabolism, drug toxicity evaluation, and hepatocellular carcinoma research.
C3A, also known as HepG2/C3A, is a clonal derivative of the human HepG2 hepatoblastoma cell line, derived from the liver tumor of a 15‑year‑old Caucasian male patient with hepatocellular carcinoma. C3A exhibits well‑differentiated epithelial morphology, adherent culture properties, and strong contact inhibition upon reaching confluence. Compared to its parental HepG2, C3A is distinguished by robust secretion of albumin and alpha‑fetoprotein (AFP), retains high glucose metabolism capability, and uniquely grows in glucose‑deficient medium. Karyotypically, it maintains aneuploid human hepatoblastoma genome typical of HepG2 derivatives. Though tumor‑derived, C3A shows limited in vivo tumorigenicity and is most valued as a stable, differentiated model of hepatic function with modest proliferation rates in vitro.

The C3A cell line finds diverse applications in cancer, toxicology, and immuno-oncology research. Its utility extends to studying hepatocyte function and targeted protein expression. Moreover, C3A cells serve as a foundation for creating sophisticated in vitro three-dimensional (3D) cell-based systems that closely mimic in vivo cellular behavior. Particularly, the utilization of HepG2/C3A spheroids presents a highly sensitive and promising model for genotoxicity assessments and environmental studies, enhancing the accuracy of evaluating genotoxic activities in both individual chemicals and intricate environmental samples.
