C3A, also known as HepG2/C3A, a derivative of Hep G2, is a hepatoblastoma cell line that was derived from the liver tissue of a 15-year-old white male patient with hepatocellular carcinoma. C3A exhibits epithelial-like morphology and adherent culture properties. These cells have the characteristics of strong contact inhibition of growth, and they are able to grow in a glucose-deficient medium. C3A expresses a high level of albumin and alpha-fetoprotein (AFP) protein, with AFP secretion decreasing and albumin secretion increasing upon cell confluence.

The C3A cell line finds diverse applications in cancer, toxicology, and immuno-oncology research. Its utility extends to studying hepatocyte function and targeted protein expression. Moreover, C3A cells serve as a foundation for creating sophisticated in vitro three-dimensional (3D) cell-based systems that closely mimic in vivo cellular behavior. Particularly, the utilization of HepG2/C3A spheroids presents a highly sensitive and promising model for genotoxicity assessments and environmental studies, enhancing the accuracy of evaluating genotoxic activities in both individual chemicals and intricate environmental samples.
