Caspase-2 is an upstream initiator of apoptosis signaling, specifically hydrolyzes the peptide substrate VDVAD-pNA (Val-Asp-Val-Ala-Asp-p-nitroanilide).
Caspases are a family of proteases involved in apoptosis, encompassing over ten members. Caspase-2, also known as Ich-1 or Nedd-2, is activated during apoptotic signaling. Caspase-2 mRNA has two distinct splice forms: the protein translated from the full-length mRNA promotes apoptosis, while the shortened protein product inhibits apoptosis. Caspase-2 can be activated by caspases 1 and 3, as well as granzyme B. Assay principle: Caspase-2 specifically hydrolyzes the peptide substrate VDVAD-pNA (Val-Asp-Val-Ala-Asp-p-nitroanilide), releasing free nitroanilide (pNA) with a maximum absorbance at 405 nm.
