Caspase-9 can initiate apoptosis. The assay is based on the specific peptide substrate LEHD-pNA (Leu-Glu-His-Asp-p-nitroanilide) hydrolysis by caspase-9.
Caspase-9, also known as ICE-LAP6 or Mch6, forms a complex with cytochrome c and Apaf1, leading to activation. This complex further activates caspase-3, a key enzyme in apoptosis, thereby triggering the apoptotic cascade. Caspase-9 is a crucial upstream caspase in apoptotic signal transduction, and its activation can be regulated by phosphorylation. The assay is based on the specific hydrolysis of the peptide substrate LEHD-pNA (Leu-Glu-His-Asp-p-nitroanilide) by caspase-9. The released free nitroanilide pNA has a maximum absorbance at 405 nm.
