Cell Senescence β-Galactosidase Staining Kit is designed based on the biological characteristics of changes in SA-β-gal activity, X-Gal is a substrate.
Cellular senescence is a regulatory mechanism that limits the proliferative capacity of cells. Commonly referred to as replicative senescence (RS), it describes the process by which normal cells permanently cease division after a finite number of cell cycles. Although senescent cells remain viable, they undergo pronounced changes in morphology and physiological metabolic activity. Two key hallmarks of cellular senescence in vitro are irreversible growth arrest and increased activity of senescence-associated β-galactosidase (SA-β-gal). SA-β-gal is a lysosomal hydrolase that is normally active at pH 4.0. However, in senescent cells, this enzyme exhibits detectable activity at the suboptimal pH of 6.0, a feature widely used as a biomarker for senescence. The proportion of cells expressing SA-β-gal increases progressively with higher passage numbers in culture.

Cell Senescence β-Galactosidase Staining Kit detects senescence-specific β-galactosidase (SA-β-Gal) in cells. It measures fluorescence intensity after cell lysis, and further detect cellular aging and longevity. It can be used in various types of cells such as adipose-derived stem cells, cancer cell lines.This kit is based on changes in SA-β-gal activity during senescence. Using X-Gal as a substrate, senescence-associated β-galactosidase catalyzes the formation of an insoluble deep blue product, allowing senescent cells to be readily visualized and analyzed under a light microscope.
