H9 cell line provides a pluripotent stem cell model for developmental biology, differentiation studies, and translational research in human cells.
H9 is a human cutaneous T-cell lymphoma line derived from Sezary syndrome (a leukemic form of mycosis fungoides), it is a derivative of the Hut 78 cell line that was originally isolated from the haematopoietic and/or lymphoid tissue of a 53-year-old white male patient. H9 exhibits lymphoblast-like morphology in suspension culture. H9 consistently expresses CD4 as well as HLA antigens A1, B62, C3, DR4, and DQ3. Karyotypic analysis reveals a near‑triploid chromosomal content, typically about 69 chromosomes (range 58–74). H9 is capable of productive HIV‑1 infection and isolation, making it valuable for virology research. Its tumorigenic nature is confirmed by subcutaneous tumor formation in immunodeficient mice. Its origin from malignant T‑cell lymphoma supports oncogenic phenotype retention and immune antigen expression consistent with cutaneous T‑cell malignancies.

H9 is widely used as a robust in vitro model for HIV‑1 infection and replication studies, particularly for evaluating virus–host interactions, viral entry, and immune modulation in CD4⁺ T‑cells. It enables detailed analysis of alterations in antigen expression and host response following infection and supports investigation of antiviral therapeutics. In addition, H9’s tumorigenic potential allows its use in in vivo xenograft studies for cutaneous T‑cell lymphoma biology and immuno-oncology research.
