Human CLDN1 Knockout A-431 Cell Line is engineered from human epidermoid carcinoma A-431 cells using CRISPR/Cas9 technology to disrupt the Claudin 1 (CLDN1) gene, a tight junction protein critical for epidermal barrier function and cell polarity. This knockout model exhibits compromised tight junction integrity and increased paracellular permeability while maintaining A-431 cells’ characteristic epithelial morphology. The cell line serves as a valuable tool for studying skin barrier disorders (such as ichthyosis and atopic dermatitis), viral entry mechanisms (including HCV and Dengue virus infection), and cancer metastasis, particularly in research involving epithelial barrier dysfunction and cell-cell adhesion. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and immunofluorescence staining demonstrating loss of tight junction strands. Rigorous quality control confirms the cell line is free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, fungi, yeast, and bacteria.
