NLRC4 Knockout THP-1 Cell Line is engineered from human monocytic THP-1 cells using CRISPR/Cas9 technology to disrupt the NOD-like receptor family CARD domain-containing protein 4 (NLRC4) gene, a critical component of inflammasome complexes that mediates caspase-1 activation and pyroptosis. This knockout model exhibits impaired inflammasome assembly and reduced IL-1β/IL-18 secretion while maintaining typical THP-1 characteristics including suspension growth with round morphology and PMA-induced macrophage differentiation capacity. The cell line serves as a valuable tool for studying intracellular pathogen recognition, inflammasome regulation, and inflammatory responses in bacterial infections and autoinflammatory disorders. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and functional assays demonstrating defective inflammasome activation. All cells are rigorously tested and confirmed free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, fungi, yeast, and bacteria.
