WM-266-4 is a human malignant melanoma cell line derived from a skin metastasis of a 58-year-old female patient with stage III (Clark level) melanoma. These adherent cells exhibit an epithelial-like, small, flat, mesenchymal morphology. WM-266-4 harbors the oncogenic BRAF V600D mutation, driving constitutive MAPK pathway activation, and displays PTEN loss of function due to hemizygous deletion. The cell line is wild-type for N-RAS, c-KIT, and CDK4, providing a distinct model to study BRAF-driven melanoma. WM-266-4 cells are tumorigenic and form xenografts in immunodeficient mice, supporting their use in melanoma progression and targeted therapy research. Their stable growth, defined genetic background, and reproducible tumorigenicity make them valuable for preclinical drug testing and mechanistic studies of metastatic melanoma.
