AceSeq™ λ Phage DNA Kit

Description

AceSeq™ λ Phage DNA Kit is suitable for rapidly extracting DNA from λ phage lysate cultures. After centrifugation of λ phage lysate culture, the supernatant is first digested with RNase A/DNase I mixed enzyme to remove the residual host bacterial DNA/RNA, and the phage is collected by precipitation. The phage is cleaved by SDS, and the residual debris is removed by precipitation centrifugation. The λ phage DNA in the supernatant of the lysate is selectively adsorbed to the silica matrix membrane in the spin column in high-salt state, and then through a series of rapid rinsing-centrifugation steps, impurities such as salt, cell metabolites, and proteins are removed. Finally, a low-salt elution buffer elutes the λ phage DNA from the silica-based membrane. The obtained λ phage vector is widely used for library screening.
