AceSeq™ DNA Phosphorylation Kit is developed for phosphorylating the -OH group at the 5’ end of DNA molecules. The 5’ ends of synthetic PCR primers, linkers and adapters are generally the -OH groups instead of the -PO4 groups. And any DNA ligase cannot combine the -OH groups at the 5’ end of one DNA molecule to the -OH group at the 3’ end of another DNA molecule. Therefore, the connection efficiency between artificially synthesized DNA fragments and natural DNA is generally lower than that of natural DNAs. This kit can quickly change the -OH group at the 5’ end of one DNA molecule to the -PO4 group, so that artificially synthesized DNA has the same high ligation efficiency as natural DNA.
