Alamar Blue is a redox indicator that undergoes measurable changes in absorbance and fluorescence in response to cellular metabolic activity. It is a safe, non-toxic dye widely used for the quantitative assessment of cell viability and proliferation, as well as for cytokine bioassays and in vitro cytotoxicity studies. The assay effectively reflects the native metabolic activity of animal, fungal, and bacterial cells. In its oxidized form, Alamar Blue appears nonfluorescent and violet-blue, while in its reduced form it is converted into a pink, fluorescent product, indicating oxygen consumption by metabolically active cells or microorganisms. These reducing equivalents convert the dye into its fluorescent form, which is released into the culture medium, causing a color change from indigo blue to pink. Cells with low metabolic activity generate weaker signals. Fluorescence can be measured using excitation wavelengths of 530–560 nm and emission at 590 nm, with signal intensity proportional to the number of active cells.

This assay is based on the ability of viable cells to generate a reducing environment during proliferation, characterized by increased ratios of intracellular redox couples such as NADH/NAD, NADPH/NADP, FADH/FAD, and FMNH/FMN.
