The Annexin V-Fluor 647/7-AAD Cell Apoptosis Detection Kit can be used to distinguish early apoptotic cells from late apoptotic or necrotic cells.
The Annexin V–Fluor 647/7-AAD Cell Apoptosis Detection Kit is designed for the detection of early apoptosis using Fluor 647–labeled Annexin V as a fluorescent probe. In viable cells, phosphatidylserine (PS) is located on the inner leaflet of the plasma membrane. During early apoptosis, PS is translocated to the outer leaflet and exposed to the extracellular environment. Annexin V is a 35–36 kDa Ca²⁺-dependent phospholipid-binding protein that binds PS with high affinity, enabling selective labeling of early apoptotic cells. When used together, live cells are Annexin V⁻/7-AAD⁻, early apoptotic cells are Annexin V⁺/7-AAD⁻, and late apoptotic or necrotic cells display double-positive staining (Annexin V⁺/7-AAD⁺).

The Annexin V-Fluor 647/7-AAD Apoptosis Detection Kit is developed to detect apoptosis in both suspension and adherent cells and is compatible with fluorescence microscopy, flow cytometry, and other fluorescence-based detection platforms.The kit also includes 7-aminoactinomycin D (7-AAD) to identify late apoptotic or necrotic cells. 7-AAD is a nucleic acid dye with fluorescence characteristics similar to propidium iodide but with a narrower emission spectrum, reducing spectral overlap in multicolor analyses. 7-AAD cannot penetrate intact cell membranes, only stains late apoptotic or necrotic cells.
