Bax Knockout 293T Cell Line is engineered from human embryonic kidney 293T cells through CRISPR/Cas9-mediated disruption of the BCL2-Associated X Protein (Bax) gene, a key pro-apoptotic regulator of mitochondrial outer membrane permeabilization and intrinsic apoptosis. This knockout model exhibits profound resistance to apoptotic stimuli while maintaining 293T cells’ characteristic adherent growth with epithelial morphology and high transfection efficiency. The cell line provides a critical tool for studying programmed cell death mechanisms, cancer therapy resistance, and mitochondrial dysfunction, particularly in research involving chemotherapeutic agents and DNA damage responses. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through multiplex verification including genomic PCR, Sanger sequencing, and functional apoptosis assays demonstrating impaired cytochrome c release.
