Hamster Syrian kidney cell line BHK 21 CL13 is susceptible to adenovirus 25, vaccinia, herpes simplex, reovirus 3, rubella, and pseudorabies viruses.
BHK-21 CL13 is a fibroblast cell line derived from baby Syrian (golden) hamster kidney tissue originally isolated in 1961 and subsequently single cell cloned to create clone 13. These cells exhibit classic spindle-shaped, adherent morphology and grow as monolayers under standard culture conditions. Chromosomal analysis reveals aneuploid karyotype with characteristic structural variations typical of immortalized BHK derivatives. They lack reverse transcriptase activity. While not inherently tumorigenic at low inoculum, BHK-21 CL13 cells can form tumors in vivo at high doses and undergo transformation upon exposure to chemical carcinogens such as MNNG or polycyclic aromatic hydrocarbons (PAHs). The cells undergo stringent screening and testing procedures to ensure they are free from contamination by Mycoplasma, fungi, yeast, and bacteria.

BHK-21 CL13 cells serve as a classic in vitro platform for viral research and are widely utilized, particularly in rabies virus diagnostics and viral susceptibility studies involving agents such as adenovirus, vaccinia, herpes simplex virus, reovirus 3, and vesicular stomatitis virus. They are also widely employed in toxicology and chemical carcinogenesis research, providing a transformation model for assessing the oncogenic potential of environmental compounds, including PAHs. Their high transfection efficiency makes them suitable for gene expression, viral propagation, and recombinant protein production.
