This knockout model exhibits impaired cell-cell adhesion and enhanced migratory capacity while maintaining 293T cells’ characteristic adherent growth (with reduced intercellular contacts) and high transfection efficiency. The cell line provides a valuable tool for studying epithelial-mesenchymal transition (EMT), cancer metastasis, and tumor microenvironment interactions, particularly in research involving Wnt/β-catenin signaling and TGF β pathways. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and functional cell aggregation assays, with additional confirmation by loss of membrane E-cadherin staining.
