CFDA, SE (carboxyfluorescein diacetate, succinimidyl ester) is a cell tracking dye that can fluorescently label living cells. It is used not only for in vitro cell proliferation experiments but also for tracking cell division and proliferation in vivo. CFDA, SE is a derivative of fluorescein diacetate (FDA). It is cell membrane-permeable and non-fluorescent. After passively transporting through the cell membrane and entering living cells, it is catalyzed by cytoplasmic esterases to form carboxyfluorescein succinimidyl ester (CFSE), which emits strong green fluorescence. CFSE is impermeable to cell membranes and remains intact within the cell. CFSE also spontaneously and irreversibly binds to amino groups within the cell, conjugating to cellular proteins. The fluorescence of cells labeled with CFDA, SE is highly uniform, surpassing that of previously used fluorescent cell-tracking probes such as PKH26. Cells labeled with CFDA, SE can be used for in vitro and in vivo proliferation studies without staining neighboring cells.

CFDA, SE is a cell-permeable dye used for cell proliferation assays, motility assays, and in vivo cell tracking experiments. CFDA, SE are most commonly used to assay the proliferation of lymphocytes, but can also be used to assay the proliferation of other cells, such as fibroblasts, natural killer cells, and hematopoietic progenitor cells.
