GH3 is a rat pituitary tumor cell line isolated from the pituitary gland of a 7-month-old female Wistar-Furth rat with a tumor. This cell line exhibits epithelial-like morphology and grows as an adherent monolayer culture. The stemline karyotype model number of GH3 is 67 with a range from 47 to 71, and two dicentric marker chromosomes were observed in all of the cells examined. The GH3 cells constitutively express prolactin and growth hormone (somatotropin). They are tumorigenic. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

GH3 serves as an in vitro model derived from a rat pituitary tumor, primarily used to investigate pituitary hormone regulation and related disorders. Its key application involves studying the secretion mechanisms, pharmacological responses, and genetic modulation of prolactin and growth hormone. This enables research into endocrine pathways, including responses to thyroid-stimulating hormone and glucocorticoids like hydrocortisone. GH3 cells are particularly valuable for drug screening assays targeting pituitary dysfunctions, providing insights for therapeutic development in endocrine diseases.
