Human EPCAM Knockout A-431 Cell Line is generated from human epidermoid carcinoma A-431 cells through CRISPR/Cas9-mediated disruption of the Epithelial Cell Adhesion Molecule (EPCAM) gene, a transmembrane glycoprotein critical for cell-cell adhesion and Wnt/β-catenin signaling. This knockout model exhibits impaired epithelial integrity and reduced proliferation while maintaining A-431 cells’ characteristic adherent growth with cobblestone morphology. The cell line serves as an essential tool for studying epithelial-mesenchymal transition (EMT), cancer stem cell biology, and targeted therapy resistance, particularly in research involving circulating tumor cells (CTCs) and antibody-based cancer therapies. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and flow cytometry confirming loss of EPCAM surface expression.
