This BAF3 cell line with ETV6-FGFR4 (C477A) mutation provides a model for studying altered kinase activity and targeted therapy development.
The ETV6 FGFR4 (C477A) BAF3 Cell Line is a genetically engineered model derived from murine BAF3 cells. It stably expresses the human ETV6-FGFR4 fusion gene harboring the C477A mutation, introduced via lentiviral transduction. This mutation resides in the FGFR4 kinase domain, disrupting cysteine-mediated disulfide bond formation and enhancing constitutive activation.
Target
ETV6-FGFR4 fusion promotes ligand-independent FGFR4 activation and is implicated in hematologic malignancies. The C477A mutation enhances kinase activity and drives oncogenic signaling pathways such as MAPK and PI3K/AKT.
