Human MMP14 Knockout A-431 Cell Line is generated from human epidermoid carcinoma A-431 cells through CRISPR/Cas9-mediated disruption of the Matrix Metalloproteinase 14 (MMP14/MT1-MMP) gene, a membrane-anchored protease critical for extracellular matrix (ECM) remodeling and tumor invasion. This knockout model exhibits impaired collagen degradation and reduced invasive capacity while maintaining A-431 cells’ characteristic epithelial morphology. The cell line provides an essential tool for studying cancer metastasis, wound healing disorders, and angiogenesis, particularly in research involving ECM degradation, invadopodia formation, and activation of other MMPs (e.g., MMP2). Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and functional gelatin degradation assays demonstrating loss of proteolytic activity. The cell line has undergone rigorous quality control and is certified free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, fungi, yeast, and bacteria.
