Human Non-Hodgkin Lymphoma (DLBCL) Peripheral Blood Mononuclear Cells (PBMCs) are isolated from patients diagnosed with diffuse large B-cell lymphoma, an aggressive form of non-Hodgkin lymphoma. They consist of lymphocytes (T cells, B cells, NK cells) and monocytes that exhibit tumor-driven immune dysregulation. These PBMCs contain malignant CD19⁺/CD20⁺ B lymphocytes alongside reactive immune populations including T cells (CD3⁺/CD4⁺/CD8⁺) and NK cells, maintaining characteristic lymphoma-associated surface markers (CD10, BCL-2, BCL-6). The cells exhibit disease-specific gene expression profiles with upregulated MYC, BCL-2, and NF-κB pathway components. DLBCL PBMCs demonstrate altered proliferative capacity and cytokine production patterns (elevated IL-10, TNF-α) compared to healthy controls. These cells serve as a valuable model for studying lymphoma-immune interactions, particularly for evaluating CAR-T cell efficacy and developing immunotherapies that target the immunosuppressive tumor microenvironment in aggressive B-cell malignancies. These circulating-associated PBMCs also provide a relevant platform for investigating DLBCL-related immune signaling features, enabling more precise modeling of immune dysregulation associated with activated B-cell–type DLBCL.
