Normal rat ileum. Derived from normal epithelial cells of the rat ileum.
IEC-18 is a rat small intestinal epithelial cell line originally derived from the ileal crypts of neonatal Rattus norvegicus (Sprague Dawley rat) and is classified as a non-transformed epithelial model. It exhibits typical epithelial morphology and grows as adherent monolayers under standard culture conditions with a doubling time of approximately 16.6 hours. IEC-18 cells form tight intercellular junctions including desmosomes and develop microvilli on their apical surface, resembling native small intestinal epithelium in structure and barrier function based on published literature characterizing epithelial features. Unlike malignant tumor lines, IEC-18 cell line is not tumorigenic in vivo. This line retains physiological properties relevant to intestinal epithelial permeability and barrier studies and has been widely adopted in research on paracellular transport, epithelial differentiation, and epithelial physiology. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

IEC-18 cells are used as an in vitro model of rat small intestinal epithelium for studies of epithelial barrier function, paracellular permeability, cell polarity, and nutrient transport mechanisms. They support investigations into tight junction regulation, ion channel activity, and responses to pharmacological or toxicological challenges relevant to intestinal physiology and drug absorption research.
