Interleukin-31 (IL-31) is a novel cytokine that belongs to the IL-6 cytokine family, with its receptor being similar to the IL-6 receptor. It is primarily secreted by activated CD4+ T lymphocytes, especially activated Th2 cells, mast cells, macrophages, and dendritic cells. IL-31 mainly regulates cell-mediated immunity in the skin by perceiving itch through the peripheral nervous system, modulates lung immunity by increasing airway inflammation, and adjusts intestinal immunity by defending against microbes.IL-31 forms a heterodimer complex signaling through the IL31RA and OSMR (oncostatin M receptor) receptors, closely associated with chronic pruritic skin diseases like atopic dermatitis. Monoclonal antibodies targeting IL-31 or its receptor can effectively reduce itching and sleep disturbances, improve skin lesions, and minimize the use of topical steroids.BA/F3 cells, dependent on IL-3 for growth, can be sustained by IL-31 protein instead of IL-3. Blocking this action with IL-31-related antagonists leads to concentration-dependent cell death.
