Description

MBT-2 is a murine bladder cancer cell line derived from a transitional cell carcinoma in the urinary bladder of a female C3H/He mouse, likely aged 6–8 weeks. Induced by FANFT, these adherent, spindle-shaped cells show fibroblast-like morphology and rapid growth (~24 h doubling time). MBT-2 is highly tumorigenic and metastatic, frequently spreading to the lungs and lymph nodes in syngeneic mice. The line is polyploid (4N–16N) and lacks p16^INK4a^ expression. MBT-2 cells express epithelial markers like cytokeratin. The cell line harbors a replicating type C retrovirus, impacting immune studies. Its robust tumorigenicity makes MBT-2 a reliable model for bladder cancer therapy and metastasis research.

Application

  • MBT-2 serves as a powerful tool for researchers to investigate the molecular mechanisms driving bladder cancer progression and metastasis. It can be grown in C3H/He mice to create a syngeneic model to test the efficacy and response to immunotherapy treatments.

Frequently Asked Questions

  • What is the culture medium of MBT-2 cell line?

Recommended Culture Medium and Conditions for MBT-2 Cells:
EMEM medium with 10% fetal bovine serum. Some data recommend the MBT-2 to culture in 1640 medium. It depends on your provider laboratory data. Antibiotics such as penicillin-streptomycin are commonly added to prevent contamination.

  • What is the doubling time of MBT-2 cell line?

The doubling time of MBT-2 cells is approximately 1 day, though this can vary depending on culture conditions. Cells are harvested after treatment with 0.25% trypsin and 0.02% EDTA.

  • What are some potential issues with the MBT-2 cell line?

In 2000, the cell line was also found to be replicating type C retrovirus contamination.
As with any cell line, there’s a risk of contamination, either with other cell lines or with microorganisms. Contamination can compromise experimental results and lead to erroneous conclusions. Ensuring that the cell line used in experiments is crucial.
