Interleukin-18 (IL-18), also known as interferon-gamma-inducing factor, is a protein encoded by the IL18 gene in the human body. This protein encoded by the gene is a pro-inflammatory cytokine. Various cell types, including hematopoietic cells and non-hematopoietic cells, have the potential to produce IL-18.Free IL-18 binds to specific heterodimer cell surface receptors, which are members of the IL-1 receptor/Toll-like receptor superfamily, consisting of two subunits, IL-18Rα (IL-18Rα1) and IL-18Rβ (IL-18RAP), both containing a Toll/IL-1R structure domain (TIR) in the cell interior. Interaction between IL-18 and IL-18Rα stabilizes its connection with IL-18Rβ through the TIR domain. This domain recruits the MyD88 adapter protein, which in turn recruits the IL-1 receptor-associated kinase (IRAK) to initiate signal transduction. IRAK gets phosphorylated and dissociates from the receptor complex, then interacts with TRAF6, ultimately activating downstream reporter genes.AcceGen’s  Mouse_IL18 Reporter 293 Cell Line, based on the reporter gene signaling pathway, is a Luciferase reporter gene cell line. When IL-18 binds to the heterodimer receptor, it recruits the MyD88 adapter protein, which in turn recruits IRAK, binds to TRAF6, activates downstream reporter genes, and thus activates the expression of luciferase. The luciferase readings represent the activation effect of the signaling pathway and can therefore be used for the in vitro evaluation of IL-18-related drugs.
