The NFAT Reporter Jurkat Cell Line is a robust tool for monitoring NFAT (Nuclear Factor of Activated T cells) signaling pathways. NFAT is a pivotal transcription factor family that drives gene expression during immune responses. Beyond T cells, NFAT is widely expressed in B cells, NK cells, mast cells, monocytes, and eosinophils, where it regulates processes such as T cell activation, differentiation, and immune tolerance. Activation of NFAT has been implicated in a broad spectrum of acute and chronic diseases, including bronchial asthma, Alzheimer’s disease, inflammatory bowel disease, diabetes, osteoporosis, osteoarthritis, and myocarditis, highlighting its critical role in immune regulation and disease pathogenesis. This reporter cell line is generated using Jurkat T cells via lentiviral transduction to stably express a luciferase reporter gene under NFAT-responsive elements. Upon stimulation with pro-inflammatory cytokines or receptor agonists, endogenous NFAT proteins bind to the DNA response elements, triggering luciferase expression. This provides a sensitive and quantitative platform for evaluating NFAT pathway activation and screening compounds that modulate NFAT signaling.
