Polybrene (Hexadimethrine Bromide) is a cationic polymer widely used to enhance the efficiency of DNA transfection in mammalian cells, particularly in conjunction with lipid-based methods and reverse or lentiviral transduction. It functions by neutralizing electrostatic repulsion between viral particles and cell surface sialic acids, thereby promoting viral adsorption and cellular uptake. Beyond virology, Polybrene serves as an effective heparin antagonist for inducing nonspecific erythrocyte agglutination. It is also employed in protein sequencing, where low concentrations significantly reduce peptide degradation during automated Edman degradation, and can improve binding affinity when applied to PVDF membranes in blotting applications.
