Raw264.7/NFκB-SEAP Reporter Cell Line is a genetically engineered macrophage model derived from murine Raw264.7 cells, designed for sensitive monitoring of NFκB (Nuclear Factor-kappa B) signaling pathway activation through secreted embryonic alkaline phosphatase (SEAP) reporter expression. This cell line maintains its monocyte/macrophage origin with characteristic adherent morphology while preserving key macrophage functions including cytokine production and phagocytic activity. The integrated NFκB-responsive SEAP reporter enables non-destructive, quantitative detection of NFκB activation through simple measurement of SEAP activity in culture supernatant, making it particularly valuable for time-course studies and high-throughput screening of anti-inflammatory compounds. Cellular markers including CD14 and F4/80 are retained, with SEAP secretion levels serving as a sensitive indicator of NFκB transcriptional activity. The cells are maintained at low passage numbers (<P20) to ensure consistent reporter response. The Raw264.7/NFκB-SEAP Reporter Cell Line provides a sensitive secreted alkaline phosphatase (SEAP) reporter system for monitoring NFκB pathway activation in macrophages.
