Description

SK-BR-3 Luciferase Reporter Cell Line is a stable, clonal derivative of the SK-BR-3 human breast cancer line, originally isolated from the pleural effusion of a 43-year-old female patient with adenocarcinoma. These cells display epithelial morphology, grow adherently as monolayers, and are defined by amplification and overexpression of the HER2 (ERBB2) oncogene, a marker of aggressive breast cancer. This reporter line has been engineered to stably express firefly luciferase, enabling constitutive bioluminescence for sensitive, real-time monitoring of cellular activity both in vitro and in vivo. This dual feature makes it a powerful model for drug screening, tumor progression studies, and investigation of HER2-driven oncogenesis and therapeutic responses.

Application

  • The SK-BR-3 Luciferase Reporter Cell Line serves as a powerful tool for HER2-positive breast cancer research, particularly in elucidating drug resistance mechanisms. These engineered cells enable researchers to investigate critical molecular pathways, providing insights into therapeutic targets and strategies to overcome resistance in this aggressive cancer subtype. The luciferase expression allows for real-time monitoring of cell viability and drug response dynamics. With its tumorigenic properties, SK-BR-3 line is valuable for establishing adenocarcinoma models in immunodeficient mice, facilitating preclinical evaluation of the antiproliferative activity of novel therapies and assessment of antiproliferative effects.

Frequently Asked Questions

  • What is the purpose of using SK-BR-3 Luciferase Reporter Cell Line?

The SK-BR-3 Luciferase Reporter Cell Line is used for studying eukaryotic gene expression and cell physiology. The luciferase gene system is applied due to its high sensitivity and success rate, enabling various applications such as promoter activity research, mammalian cell two-hybrid experiments, and in vivo imaging experiments.

  • How is luciferase activity validated?

Luciferase activity is validated using the Dual-Luciferase® Reporter Assay System Kit. The firefly luciferase reaction intensity is measured using a microplate reader, ensuring accurate detection of luciferase activity.

  • What is the recommended procedure to thaw SK-BR-3-Luc cells?

    To thaw SK-BR-3-Luc cells:
Warm the complete culture medium to 37°C.
Transfer the cryopreserved vial from liquid nitrogen to a -80°C freezer briefly, then thaw it in a 37°C water bath.
After thawing, centrifuge the cells, discard the supernatant, and resuspend them in fresh medium before incubating them in a T25 flask in a 37°C, 5% CO₂ incubator.

  • What are the shipping and storage conditions for the SK-BR-3 Luciferase Reporter Cell Line?

The cells are shipped on dry ice. Upon receipt, they should be transferred immediately to a liquid nitrogen tank (-180°C) for long-term storage. The cells should not be stored at -20°C as this will not preserve them properly.

  • What quality control measures are implemented for the SK-BR-3-Luc Cell Line?

Each vial of the SK-BR-3-Luc cell line contains over 1×10⁶ cells with more than 90% viability. The cells are tested negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
