THP-1/GFP-Luciferase Stable Cell Line is a dual-reporter human monocytic cell line engineered from THP-1 cells through stable co-expression of green fluorescent protein (GFP) and luciferase (Luc), the cell line maintains key native THP-1 characteristics, such as the capacity to upregulate CD14 upon differentiation and PMA-induced differentiation into macrophage-like cells. This versatile reporter system combines the advantages of fluorescent visualization for cellular localization studies with sensitive bioluminescent detection for quantitative cell tracking, making it particularly valuable for immunological research, cancer microenvironment studies, and cell therapy development. The GFP marker enables real-time monitoring of cell migration and interactions through fluorescence imaging, while the luciferase activity permits highly sensitive in vitro and in vivo quantification of cell proliferation and viability using bioluminescence imaging (BLI) technology. Cells are maintained at low passage numbers (<P20) to ensure stable expression of both reporters and consistent experimental performance. The cells are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, Fungi, Yeast and Bacteria.

This system combines the advantages of fluorescence microscopy (GFP) for cellular localization with sensitive bioluminescence imaging (luciferase) for quantitative cell number assessment, making it ideal for studying monocyte migration, immune responses, and cell therapy applications. The cell line maintains native THP-1 differentiation capacity into macrophages while providing consistent dual-reporter expression. Maintain in standard RPMI-1640 medium supplemented with 10% FBS and appropriate selection antibiotics to ensure stable GFP and luciferase expression throughout experiments.
