THP-1/STAT5 Reporter (GFP) Stable Cell Line is a genetically modified human monocytic cell line designed to monitor STAT5 (Signal Transducer and Activator of Transcription 5) signaling pathway activation through green fluorescent protein (GFP) expression. Derived from the THP-1 parental cell line, the cell line maintains key native THP-1 characteristics, such as the capacity to upregulate CD14 upon differentiation and PMA-induced differentiation into macrophage-like cells. The cell line features a GFP reporter gene under the control of STAT5-responsive elements, enabling real-time visualization and quantitative analysis of STAT5 activation in response to cytokine stimulation (including IL-2, IL-7, and GM-CSF). With stable GFP expression maintained at low passage numbers (<P20), this cell line serves as a valuable tool for studying immune cell signaling, hematopoiesis, and potential therapeutic interventions. The cells are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, Fungi, Yeast and Bacteria.

The THP-1/STAT5 Reporter (GFP) Stable Cell Line is a human monocytic reporter system designed for real-time monitoring of STAT5 signaling pathway activation. This cell line expresses GFP under the control of STAT5-responsive elements, enabling fluorescence-based quantification of STAT5 activity in response to cytokine stimulation (e.g., IL-2, GM-CSF) or pharmacological modulation. The reporter system provides dynamic, non-destructive readouts for studying hematopoiesis, immune cell differentiation, and JAK-STAT signaling disorders. Maintain in standard RPMI-1640 medium supplemented with 10% FBS and appropriate selection antibiotics to ensure stable GFP reporter expression throughout experiments.

Yes. The parental THP-1 cells are reported to endogenously express JAK1, JAK2, TYK2, STAT5A, and STAT5B. In this reporter cell line, no exogenous JAK or STAT5 coding sequences were introduced; only a STAT5-responsive element driving GFP was introduced to enable a JAK-STAT5 signaling reporter assay. No exogenous JAK/STAT5 mutations were introduced, although the endogenous JAK/STAT5 genes have not been specifically sequenced in this cell line, so naturally occurring variants cannot be excluded.
