This Tie2 stable line is adherent, like parental HEK-293 cells. No special coating is required under routine culture. For certain experiments (e.g., ELISA or phosphorylation assays), surface coatings such as poly-L-lysine may be used to improve consistency, but pre-testing is recommended.
Culture in DMEM + 10% FBS + 1% P.S + 0.75 µg/mL puromycin. Continuous selection with puromycin is recommended to maintain stable Tie2 expression. During 1–2 passages before functional assays, puromycin can be reduced or removed to avoid potential effects on cell physiology.
Tie2 surface expression has been confirmed via flow cytometry.
Systematic validation of Tie2 phosphorylation upon Ang1/Ang2 stimulation or downstream signaling has not yet been performed for this line.
Supplied cells are Pn+5; expression is expected to be stable for over 20 passages. COA documents include STR profiling, mycoplasma testing, and post-thaw viability. Low-passage cells should be frozen for long-term use.
