BCECF (2',7' -bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein) is a pH fluorescence probe, applicable in a cellular experiments involving pH changes.
BCECF is a ratiometric pH fluorescence probe with an Ex = 503 nm and an Em = 530 nm. Combined with modern fluorescence imaging techniques, it is widely applicable in a variety of experiments involving pH changes, such as cell growth regulation, calcium regulation, receptor-mediated signal transduction, and iron ion transport and endocytosis.
BCECF, AM is based on BCECF but incorporates an acetylmethoxymethyl (AM) group, increasing its hydrophobicity and enabling it to easily penetrate living cell membranes. BCECF, AM itself is non-fluorescent. After penetrating the cell membrane, it is cleaved by intracellular esterases to form BCECF, which is retained within the cell and emits green fluorescence in response to pH changes.

BCECF-AM is a cell-permeant pH indicator in its acetoxymethyl ester (AM) form with a pKa (6.97). This form of the pH indicator passively crosses the intact, live cell membrane where the AM ester moiety is cleaved by intracellular esterases, retaining the probe inside.
