The Detroit 551 cell line is a human fibroblast model used to study the cell proliferation, gene expression, and molecular signaling in vitro.
Detroit 551 is a human skin fibroblast line derived from a skin biopsy of a normal Caucasian female embryo. These cells exhibit a spindle-shaped, elongated morphology typical of fibroblasts, with a monolayer growth pattern and prominent cytoplasmic extensions. They are mesenchymal in origin and function primarily in extracellular matrix (ECM) synthesis, including collagen and elastin production, contributing to skin structure and wound healing. Key markers for Detroit 551 include vimentin (a fibroblast intermediate filament protein) and prolyl-4-hydroxylase (involved in collagen biosynthesis). Each lot undergoes rigorous screening and isolation procedures, and is rigorously tested to ensure it is free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

Detroit 551 is widely used in toxicological, dermatological, and fibrosis-related research. One key application is in cytotoxicity testing, where it serves as a model to evaluate the effects of chemicals such as benzoic acid. Additionally, Detroit 551 fibroblasts are employed in fibrosis studies, particularly to model excessive collagen deposition and tissue scarring observed in conditions such as systemic sclerosis (scleroderma).
