EL4 ,Mouse lymphoma cell line resistant to cortisol, producing IL-2, applied in immunology, oncology, T-cell signaling, and therapy research.
EL4 is a murine T-cell lymphoma cell line established from a lymphoma induced in a C57BL mouse by 9,10-dimethyl-1,2-benzanthracene. EL4 exhibits a lymphoblast-like morphology, growing as free-floating round cells and exclusively in suspension culture. The EL4 cell line has a near-diploid chromosome count of 2n=39. The EL4 cell line expresses the H-2b and the Thy-1.2 cell surface glycoprotein. It is highly tumorigenic and exhibits strong metastatic potential. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

EL4 cells serve as a critical in vitro model system for immunotherapy research, particularly in evaluating T cell-mediated anti-tumor responses. For instance, they are extensively used in CAR-T cell development and efficacy testing using engineered variants. EL4 cells are a well-established platform for gene editing and functional genomics studies, aiding mechanistic studies in cancer immunology and lymphocyte activation.

EL4 cells are NK-T cell tumors that may have a genetic mechanism that allows them to express a limited group of molecules involved in recognition and signaling. They are small, rounded, and can be oval or multiform, and they often grow singly in suspension. Some cells may have small hairs or fringes.
EL4 cells are resistant to 0.1 mM cortisol and sensitive to 20 μg/mL PHA. The cells have been reported to produce low levels of type G (Gross) virus antigen H-2b, IL-4 and Thy-1.2 antigen. They can also continuously produce exosomes that carry TRAIL protein, which can induce apoptosis and necrosis in 4T1 cells.
The doubling time of EL4 cells is approximately 12-20 hours, though this can vary depending on culture conditions.
