This subline of the EL4 cell line is capable of producing IL-2 in response to phorbol-12-myristate-13-acetate (PMA).
EL4.IL-2 is a genetically engineered subline of the EL4 lymphoma cell line, originally isolated from the thymus of a mouse with lymphoma. This subline was created by stable transfection with a plasmid expressing murine Interleukin-2 (IL-2). EL4.IL-2 exhibits a lymphoblast-like morphology as round cells and grows exclusively in suspension culture. The cells express high levels of IL-2. EL4.IL-2 retains tumorigenicity, but its metastatic capacity is significantly reduced compared to the parental EL4 cell line in immunocompetent syngeneic mice. This reduction is attributed to enhanced immune surveillance triggered by IL-2 secretion. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

EL4.IL-2 cells serve as a valuable tool for immunological research and cancer immunotherapy development. These cells can be used as an in vitro model to study T-cell activation mechanisms and cytokine production, particularly interleukin-2 (IL-2) secretion. This application is relevant for drug screening of immunomodulatory compounds and for investigating cancer immunotherapy strategies targeting T-cell signaling pathways. Furthermore, their suitability for 3D cell culture enables more physiologically relevant studies of tumor-immune interactions.
