The FaDu line was established in 1968 from a punch biopsy of an hypopharyngeal tumor removed from a Hindu patient.
FaDu is a human hypopharyngeal squamous cell carcinoma (SCC) cell line originally isolated from a metastatic lesion of a 56-year-old male patient. This adherent epithelial line exhibits typical cobblestone morphology and forms a rapidly proliferating adherent monolayer in vitro. The FaDu cell line exhibits a chromosome number ranging from hypodiploid to hypertriploid, with a modal number of 64. FaDu cells harbor mutations in CDKN2A and FAT1, and express EGFR. They are tumorigenic in nude mice, forming well-differentiated grade I epidermoid carcinoma. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

FaDu cells serve as a critical in vitro model for studying hypopharyngeal squamous cell carcinoma. Their primary practical applications encompass cancer research into tumor biology and treatment resistance mechanisms, drug screening for evaluating novel chemotherapeutic agents and targeted therapies, immuno-oncology research investigating tumor-immune interactions and immunotherapies, and serving as a suitable transfection host for gene editing applications, including CRISPR-based functional genomics screens.
