CMFDA is a stable green fluorescent cell tracking dye enabling long term labeling, strong intracellular retention, and co staining with RFP.
CMFDA, or 5-chloromethylfluoresceindiacetate, is a fluorescent dye that freely permeates living cell membranes for cell tracking. CMFDA is a chloromethyl derivative of fluorescein diacetate (FDA). After passively crossing the cell membrane and entering living cells, the lipophilic group in CMFDA is hydrolyzed by nonspecific esterases in the cytoplasm to form 5-chloromethylfluorescein. Cells labeled with CMFDA exhibit very stable fluorescence, lasting for several days (at least three days), making it ideal for studying cell differentiation and morphogenesis. This product possesses the ideal properties of a cell-tracking reagent: stability, non-toxicity (within the working concentration range), good intracellular fluorescence retention, and strong fluorescence (within the physiological pH range). CMFDA-labeled cells exhibit green fluorescence (Ex = 492 nm, Em = 517 nm), and can be co-stained with other fluorescent dyes, such as red fluorescent protein (RFP).
