Human Asthma Peripheral Blood Mononuclear Cells (PBMCs) are mainly derived from the peripheral blood of asthma patients, separated by density gradient centrifugation, and are rich in lymphocytes (T cells, B cells, NK cells), monocytes, and dendritic cells. Typical mononuclear morphology—round lymphocytes, monocytes with kidney-shaped nuclei PBMCs of asthma patients play a central role in the Th2 immune response, which is characterized by increased secretion of cytokines such as IL-4, IL-5, and IL-13, driving eosinophilic inflammation, IgE synthesis, and airway hyperresponsiveness. Upregulated Th2 cytokines (IL-4, IL-5, IL-13), possible increased expression of activation markers like CD69 on T cells. Functional abnormalities include Th2 polarization leading to eosinophilic inflammation, increased IgE production, and airway hyperresponsiveness typical of asthma pathophysiology. In translational research, these samples are applied to evaluate the asthmatic immune response, investigate adaptive immunity in asthma, perform detailed PBMC cytokine profiling asthma, and study the overall asthma immune system response to better understand disease mechanisms and guide therapeutic development.
