Peripheral blood mononuclear cells are isolated from Rheumatoid Arthritis (RA) peripheral blood by diluting the whole blood with PBS and using gradient separation techniques. After centrifugation,the mononuclear cell layer is collected. Mononuclear Cells can be processed further to isolate subpopulations. Fresh RA PBMCs is available upon request.

Description

Human RA Peripheral Blood Mononuclear Cells (PBMCs) are isolated from the peripheral blood of patients with rheumatoid arthritis (RA). Small, round mononuclear cells typical of PBMCs, suspension growth.They contain lymphocytes (T cells, B cells, NK cells) and monocytes. PBMCs play a crucial role in immune regulation. T/B cells mediate adaptive immune responses, and monocytes can differentiate into macrophages or dendritic cells to participate in antigen presentation and inflammatory responses. Elevated activation markers on T cells (e.g., CD69, HLA-DR), increased proinflammatory cytokines such as TNF-α, IL-6, IL-17 expression. In RA, these cells are abnormally activated, leading to the production of autoantibodies (such as anti-CCP antibodies) and increased secretion of proinflammatory cytokines (TNF-α, IL-6, IL-17), driving synovial inflammation and joint destruction .Abnormal activation and autoantibody production (anti-CCP), contributing to RA pathogenesis.
