Human RA Peripheral Blood Mononuclear Cells (PBMCs) are isolated from the peripheral blood of patients with rheumatoid arthritis (RA). They contain lymphocytes (T cells, B cells, NK cells) and monocytes. Morphologically, they exihibit small, round mononuclear cell morphology and suspension growth pattern, typical for PBMCs. These PBMCs play a vital role in immune regulation with T cells and B cells mediate adaptive immunity, while monocytes can differentiate into macrophages or dendritic cells, contributing to antigen presentation and inflammatory responses. In RA, these cells are abnormally activated, leading to the production of autoantibodies (such as anti-CCP antibodies) and increased secretion of proinflammatory cytokines (TNF-α, IL-6, IL-17), driving synovial inflammation and joint destruction. The abnormal activation and autoantibody production (anti-CCP) contribute to RA pathogenesis. In translational and clinical studies, these samples are widely used to evaluate the immune system response in RA, identify joint inflammation blood markers, support the development of novel therapies involving PBMC RA, elucidate adaptive immunity in rheumatoid arthritis, and monitor RA disease progression.
