JC-1 dye accumulates within mitochondria and can be used to monitor mitochondrial membrane potential in cells, tissues, or purified samples.
JC-1 is an ideal fluorescent probe widely used to measure mitochondrial membrane potential (ΔΨm). JC-1 dye accumulates within mitochondria in a potential-dependent manner and can be used to monitor mitochondrial membrane potential in cells, tissues, or purified samples. In normal mitochondria, JC-1 aggregates within the mitochondrial matrix, forming polymers that emit strong red fluorescence (Ex=585 nm, Em=590 nm). In unhealthy mitochondria, due to decreased or absent membrane potential, JC-1 is confined to the cytoplasm as monomers, producing green fluorescence (Ex=514 nm, Em=529 nm). The commonly used concentration range for JC-1 measurement of mitochondrial membrane potential is 1-20 µg/mL, with 10 µg/mL being an appropriate concentration for many cell types.

JC-1 is a fluorescent dye used in cell biology to measure mitochondrial membrane potential, a key indicator of mitochondrial health and function. JC-1 can be used for qualitative detection, as color changes directly reflect changes in mitochondrial membrane potential. It can also be used for quantitative analysis, as the degree of mitochondrial depolarization can be measured by the ratio of red to green fluorescence intensities. Standard settings for red and green fluorescence are sufficient for observation.
