The JC-1 Mitochondrial Membrane Potential Assay Kit uses JC-1 as a fluorescent probe for rapid and sensitive detection of changes in mitochondrial membrane potential (ΔΨm) in cells, tissues, or purified samples. It is widely applied for the early detection of apoptosis, as mitochondrial membrane depolarization is a key early apoptotic event.
JC-1 accumulates in mitochondria in a membrane potential–dependent manner. In healthy mitochondria with intact ΔΨm, JC-1 forms aggregates within the mitochondrial matrix, emitting strong red fluorescence (Ex = 585 nm, Em = 590 nm). When mitochondrial membrane potential decreases or collapses, JC-1 remains in its monomeric form in the cytoplasm and emits green fluorescence (Ex = 514 nm, Em = 529 nm). The shift from red to green fluorescence allows both qualitative visualization and quantitative analysis of mitochondrial depolarization by calculating the red-to-green fluorescence intensity ratio.

JC-1 is a fluorescent dye used in cell biology to measure mitochondrial membrane potential, a key indicator of mitochondrial health and function. Loss of mitochondrial membrane potential increases membrane permeability, leading to the release of apoptogenic factors such as cytochrome C, Smac/DIABLO, HtrA2/OMI, EndoG, and AIF into the cytoplasm, thereby activating downstream apoptotic signaling cascades.
