The JC-10 Mitochondrial Membrane Potential Assay Kit use JC-10, an upgraded fluorescent probe to detect changes in mitochondrial membrane potential.
JC-10 is a mitochondrial membrane potential–sensitive fluorescent probe and an improved alternative to JC-1 for detecting changes in mitochondrial membrane potential (ΔΨm). Although JC-1 is widely used, its poor water solubility can cause precipitation in aqueous buffers even at low concentrations, limiting its application. JC-10 exhibits enhanced water solubility and can therefore replace JC-1 in experiments requiring higher dye concentrations. While JC-10 shows superior performance in some cell lines, its effectiveness remains highly cell-type dependent.
In healthy cells, JC-10 selectively accumulates in the mitochondrial matrix and forms reversible aggregates that emit red fluorescence (Ex = 540 nm, Em = 590 nm). When mitochondrial membrane potential decreases or collapses, JC-10 dissociates into monomers that localize in the cytoplasm and emit green fluorescence (Ex = 490 nm, Em = 525 nm).

JC-10 can be used to qualitatively monitor and/or visualize mitochondrial membrane potential in live cells as a static endpoint or over time.This fluorescence shift enables both qualitative visualization and quantitative assessment of mitochondrial depolarization by calculating the red-to-green fluorescence intensity ratio. The red and green signals can be detected using standard flow cytometry channels (FL2 and FL1, respectively), as well as fluorescence microscopy and microplate-based assays. The kit includes CCCP as a positive control and supports analysis of up to 100 samples using six-well plate formats.
