Description

RAW-264.7
RAW 264.7 is a murine macrophage-like cell line derived from a tumor induced by the Abelson leukemia virus in a male BALB/c mouse. These cells exhibit macrophage-like morphology with a distinctive fusiform and polygonal appearance and semi-adherent growth, often forming loose clusters in culture. The karyotype of RAW 264.7 is aneuploid, with a chromosome count of 60 to 66, placing it in the near-triploid range. RAW 264.7 cells express macrophage-specific surface markers such as F4/80 and CD14 and produce high levels of pro-inflammatory cytokines, including TNF-α, IL-1β, in response to stimulation. RAW 264.7 is non-tumorigenic in nude mice. However, it may form transient tumors in certain immunocompromised models at high inoculation doses.

Application

  • The RAW-264.7 cell line is a widely used model for studying mouse macrophage biology, particularly in immunology and microbiology research. These cells are instrumental in investigating cellular responses to microbial components, inflammation, and immune signaling. This cell line is also extensively used in osteoclast research, as RAW 264.7 cells can differentiate into osteoclast-like cells (OCLs), providing a reliable in vitro system for studying bone metabolism, osteoclastogenesis, and precursor cell migration toward bone. Additionally, RAW-264.7 cells are a preferred choice as a transfection host due to their suitability, making them a versatile tool for exploring gene expression, signal transduction, and host-pathogen interactions across diverse experimental applications.

Frequently Asked Questions

  • What is the morphology of RAW-264.7 cells?

The RAW 264.7 cells are monocyte/macrophage-like cells. The normal cell morphology generally showed a round and smooth form. A small percentage (<10%) of spindle-shaped or polygonal differentiated cells is considered normal.

  • Do you have the subculture tips for RAW-264.7 cells?

Please subculture when density reached 70-80%. Cells should not overgrow and become confluent as this can lead to loss of the flattened adherent cell characteristic, prone to differentiate small tentacles.

Cells are semi-adherent, it is recommended to use cell scrapers to gently remove attached cells. Do not use trypsin. Please seed the undifferentiated cells only.

The RAW-264.7 cell seeding is approximate 2-4×10,000 cells/cm². Unlike other cell lines, RAW-264.7 cells do not connect when density is low, instead, they tend to grow in clusters, distributed in patches in the culture vessel.

  • What is the RAW-264.7 cell differentiation?

Induction time and cell density are two key factors related to the differentiation of RAW264.7 cells. It is hard to avoid the differentiation and please control the differentiation ratio into a certain range. Please culture the RAW-264.7 cell line carefully.

Also, RAW264.7 cells might act as osteoclast progenitors and could differentiate to osteoclasts in response to stimulation substances like LPS.

  • How often should I refresh the medium and subculture the RAW-264.7 cells?

The growth media should be refreshed every 2-3 days, mostly not over 4 days. The average doubling time of RAW cells is 15 hours.
