THP-1/NFκB & IRF Reporter Cell Line by AcceGen enables dual-pathway monitoring of innate immunity via NFκB-SEAP and IRF-Lucia bioluminescent assays.
THP-1/NFκB & IRF Reporter Cell Line is a dual-reporter human monocytic model derived from THP-1 cells, engineered to monitor NFκB (nuclear factor kappa-light-chain-enhancer of activated B cells) and IRF (interferon regulatory factor) signaling pathways simultaneously. This cell line expresses secreted alkaline phosphatase (SEAP) under NFκB response elements and Lucia luciferase under IRF-responsive promoters, allowing sensitive and quantitative dual readouts of inflammatory and interferon signaling. It retains key THP-1 features, including CD14 upregulation upon differentiation and PMA-induced macrophage-like conversion, making it particularly valuable for studies of innate immune activation, pathogen recognition, vaccine adjuvant screening, and immunomodulatory drug development. Cells are cryopreserved at low passage (<P20) to ensure stable reporter performance and reproducibility. All batches are rigorously tested and confirmed free of HIV-1, HBV, HCV, syphilis, mycoplasma, fungi, yeast, and bacteria.

The THP-1/NFκB & IRF Reporter Cell Line is a dual-pathway human monocytic model engineered to simultaneously monitor both NFκB and IRF signaling activation upon TLR or cytokine stimulation. This system enables parallel quantification of inflammatory (NFκB) and interferon (IRF) responses through dual-reporter outputs, making it ideal for studying innate immunity crosstalk, vaccine adjuvant screening, and autoimmune/inflammatory disease mechanisms. Maintain in RPMI-1640 with 10% FBS and selection antibiotics to ensure stable reporter activity during THP-1 differentiation and stimulation experiments.
